    Willingness to Pay for the Maintenance of Green Infrastructure in Six Chinese Pilot Sponge Cities

    Due to the increasingly devastating impact of pluvial flooding on human beings' lives and properties in cities, the use of green infrastructure to manage stormwater onsite is becoming more popular worldwide. The maintenance of green infrastructure to ensure its function has become one of the most pressing tasks facing policy makers. However, there is limited research regarding the willingness to pay the stormwater fee as a form of maintenance funding. This study utilized contingent valuation data obtained from a survey of 1101 respondents living in six pilot sponge cities in China to estimate the willingness to pay for the green infrastructure maintenance. The findings indicated that two-thirds of all respondents were willing to pay, 17% would like to pay around 6-10 RMB/month (0.95-1.59 US dollars/month), and 17.8% would like to pay more than 20 RMB/month (3.2 US dollars/month). The educational level and age of the respondents were significant determinants of the probability of willingness to pay and the amount they would like to pay. Knowledge of the concept of sponge cities was another significant influencing factor for the willingness to pay, but it did not influence the amount of payment. The findings could help policy makers make better strategies regarding the maintenance of green infrastructure and its costs

    Citizen Willingness to Pay for the Implementation of Urban Green Infrastructure in the Pilot Sponge Cities in China

    Urban green infrastructure has been widely used to in cities to solve stormwater problems caused by extreme weather events and urbanization around the world. However, the lack of a long-term funding mechanism for performing urban green infrastructure's functions has limited wider implementation. Factors influencing citizen attitudes and willingness to pay for urban green infrastructure vary from city to city. This study estimated the public's willingness to pay for urban green infrastructure, as well as compared the selected influencing factors of willingness to pay in different Chinese pilot sponge cities. The results show that 60% to 75% of all respondents in the cities were willing to support the implementation of urban green infrastructure in sponge cities, with those most willing to pay around 0-5 RMB/month (0-0.72 USD/month). The respondents' educational level was a significant influencing factor for their willingness to pay in all six cities, but age, gender and family monthly income correlated differently with respondents' willingness to pay in different cities. Previous knowledge of the sponge city concept and sponge city construction in the community were not significantly correlated with residents' willingness to pay. We conclude that local governments in China need to provide more information to the general public about the multiple ecosystem services, e.g., educational and recreational benefits, that urban green infrastructure can provide. In doing so, it will help a shift to urban green infrastructure as the solution to dealing with urban stormwater problems

    DprA Is Essential for Natural Competence in Riemerella anatipestifer and Has a Conserved Evolutionary Mechanism

    Riemerella anatipestifer ATCC11845 (RA ATCC11845) is naturally competent. However, the genes involved in natural transformation in this species remain largely unknown. Bioinformatic analysis predicts that DprA of RA (DprARa) has three domains: a sterile alpha motif (SAM), a Rossmann fold (RF) domain and a Z-DNA-binding domain (Zα). Inactivation of dprA abrogated natural transformation in RA ATCC11845, and this effect was restored by the expression of dprA in trans. The dprA with SAM and RF domains of Streptococcus pneumoniae and the dprA with RF and Zα domains of Helicobacter pylori was able to restore natural transformation in the RA ATCC11845 dprA mutant. An Arg123 mutation in the RF domain of R. anatipestifer was not able to restore natural transformation of the RA ATCC11845 dprA mutant. Furthermore, DprAR123E abolished its ability to bind DNA, suggesting that the RF domain is essential for the function of DprA. Finally, the dprA of Fusobacterium naviforme which has not been reported to be natural competent currently was partially able to restore natural transformation in RA ATCC11845 dprA mutant. These results collectively suggest that DprA has a conserved evolutionary mechanism

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Urban sustainable stormwater management described from a governance perspective – challenges and interdependencies

    Conventional stormwater management systems have been criticised as being unsustainable due to increased flood risks downstream and water quality degradation in the receiving water bodies. Use of green infrastructure to manage stormwater has long been suggested, but implementation is still mainly limited to pilot projects. Based on a systematic literature review and two qualitative case studies, governance challenges and their interdependencies influencing implementation of sustainable stormwater management (SSM) were examined. In the literature review, nineteen governance challenges hindering SSM implementation from a global perspective were identified. Case study 1 examined governance factors influencing SSM implementation from a local perspective under two distinct governance structures, namely, hierarchical in China and non-hierarchical in Sweden. Governance factors found to influence SSM implementation were mapped into causal loop diagrams (CLD) to describe the governance factor interdependencies. Case study 2, investigating the Sponge Cities initiative in China, identified governance challenges influencing a national SSM policy being implemented locally. It was found that conventional grey infrastructure still plays a significant role in the Sponge Cities initiative due to lack of space, a general lack of knowledge of SSM and prioritization of quantifiable objectives within a short time-frame. The thesis provides an overview of governance challenges and their interdependencies influencing SSM implementation, as well as a visual tool (SSM-CLDs) to help local authorities and non-governmental stakeholders understand the complexities of SSM from a governance perspective

    Evaluating the Landscape Quality of Residential Communities: A Case Study of the Chinese City Yangling

    Due to continuous urbanization and an increasing need to improve living quality, citizens’ pursuit of landscape quality in residential communities is constantly improving in developing countries, e.g., China. This is especially true in the period when citizens were locked down in their home cities or communities in the context of the COVID-19 pandemic. Studying whether the current landscape in residential communities still meets citizens’ needs is of significance as it is crucial for city planners, landscape architects, and city managers. In this study, we used the analytic hierarchy process method to evaluate the landscape quality of five residential communities, using the case city of Yangling, China. In total, 516 valid questionnaires were collected in May 2022. The results showed that good organization of residential roads and pedestrian systems, the rationality of rest facilities, and the hierarchical richness of plants were the most important aspects for residents. Based on these, optimization design strategies were summarized. We hope to provide a reference for future landscape optimization of existing residential communities, especially in developing countries

    Exosomal MicroRNA MiR-1246 Promotes Cell Proliferation, Invasion and Drug Resistance by Targeting CCNG2 in Breast Cancer

    Background/Aims: Treatment of breast cancer remains a clinical challenge. This study aims to validate exosomal microRNA-1246 (miR-1246) as a serum biomarker for breast cancer and understand the underlying mechanism in breast cancer progression. Methods: The expression levels of endogenous and exosomal miRNAs were examined by real time PCR, and the expression level of the target protein was detected by western blot. Scanning electron and confocal microscopy were used to characterize exosomes and to study their uptake and transfer. Luciferase reporter plasmids and its mutant were used to confirm direct targeting. Furthermore, the functional significance of exosomal miR-1246 was estimated by invasion assay and cell viability assay. Results: In this study, we demonstrate that exosomes carrying microRNA can be transferred among different cell lines through direct uptake. miR-1246 is highly expressed in metastatic breast cancer MDA-MB-231 cells compared to non-metastatic breast cancer cells or non-malignant breast cells. Moreover, miR-1246 can suppress the expression level of its target gene, Cyclin-G2 (CCNG2), indicating its functional significance. Finally, treatment with exosomes derived from MDA-MB-231 cells could enhance the viability, migration and chemotherapy resistance of non-malignant HMLE cells. Conclusions: Together, our results support an important role of exosomes and exosomal miRNAs in regulating breast tumor progression, which highlights their potential for applications in miRNA-based therapeutics
